Desc1 :
Met, a high affinity receptor for hepatocyte growth factor (HGF; also known as scatter factor), is a disulfide-linked heterodimer made of 45kD alpha and 145kD beta-subunits (1,2). The alpha-subunit and the amino-terminal region of beta-subunit form the extracellular domain. The remainder of the beta-chain spans the plasma membrane and contains a cytoplasmic region with tyrosine kinase activity. Interaction of Met with HGF results in autophosphorylation at multiple tyrosines, which recruit several downstream signaling components, including Gab1, c-Cbl and PI3 kinase (3) and is critical for all of its known biological functions. Phosphorylation of Tyr1234/1235 in the Met kinase domain is critical to kinase activation. Phosphorylation of Tyr1349 in the Met cytoplasmic domain provides a direct binding site for Gab1 (4). Altered Met levels and/or tyrosine kinase activities were found in different tumors, especially in renal, colon and breast cancers. Thus, Met is an attractive therapeutic target and cancer diagnostic (5). Applications: Suitable for use in ELISA, Western Blot, Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight. Immunoprecipitation: 1:50 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Hu Mk Mo Rt
Immunogen :
Synthetic phosphopeptide (KLH coupled) corresponding to residues surrounding Tyr1234/1235 of human Met.
Specificity :
Recognizes human Met only when phosphorylated at tyrosine 1234/1235 (Mr 145kD). This antibody crossreacts slightly with activated PDGF, insulin and FGF receptors, but it does not crossreact with other tyrosine-phosphorylated proteins. Species Crossreactivity: mouse, rat and monkey.
Purity :
Purified by Protein A and peptide affinity chromatography.
Form :
Supplied as a liquid in 10mM sodium HEPES, pH 7.5, 150mM sodium chloride, 0.1mg/ml BSA, 50% glycerol.
Concentration :
Not determined
