Desc1 :

MDM2, a ubiquitin ligase for p53, plays a central role in regulation of the stability of p53 (1) via Akt-mediated MDM2 phosphorylation at Ser166 and Ser186 (2,3). Phosphorylation of MDM2 increases its interaction with p300, providing a platform to allow the assembly of the protein complex necessary for MDM2-mediated ubiquitination and degradation of p53 (4). Phosphorylation of MDM2 also blocks its binding to p19ARF, increasing the degradation of p53 (3). Specificity/Sensitivity: Phospho-MDM2 (Ser166) antibody detects endogenous levels of MDM2 only when phosphorylated at serine 166. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
