Macrophage-colony stimulating factor (M-CSF, CSF-1) receptor is an integral membrane tyrosine kinase encoded by the c-fms proto-oncogene. MCSF receptor is expressed in monocytes (macrophages and their progenitors) and drives growth and development of this blood cell lineage. Binding of M-CSF to its receptor induces receptor dimerization, activation and autophosphorylation of cytoplasmic tyrosine residues used as docking sites for SH2-containing signaling proteins. There are at least five major tyrosine autophosphorylation sites. Tyrosine 723 (Tyr721 in mouse) is located in the kinase insert (KI) region. Phosphorylated tyrosine 723 binds the p85 subunit of PI3 kinase as well as PLC-gamma2. Phosphorylation of Tyr809 provides a docking site for Shc. Overactivation of this receptor can lead to a malignant phenotype in various cell systems. The activated MCSF receptor has been shown to be a predictor of poor outcome in advanced epithelial ovarian carcinoma and breast cancer.
Applications:
Suitable for use in Western Blot, Immunocytochemistry and ELISA. Other applications not tested.
Recommended Dilution:
Western Blot: 0.1ug/ml Detects mouse M-CSFR
Immunocytochemistry: 5-15ug/ml in immersion fixed mouse splenocytes
Optimal dilutions to be determined by the researcher.
Storage and Stability:
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
