LEF1/TCF are members of the high mobility group (HMG) DNA binding protein family of transcription factors which consists of four members: Lymphoid enhancer factor 1 (LEF1), T Cell Factor 1 (TCF1), TCF3 and TCF4 (1). LEF1 and TCF1 were originally identified as important factors regulating early lymphoid development (2) and are downstream factors in the Wnt signaling pathway. LEF1/TCF bind to Wnt response elements to provide a docking site for beta-catenin which translocates to the nucleus and promotes the transcription of target genes upon activation of Wnt signaling (3). LEF1/TCF proteins are dynamically expressed during development and aberrant activation of the Wnt signaling pathway is involved in many types of cancers such as colon cancers (4,5).
LEF1 has several isoforms due to alternative splicing. LEF1 also has an alternative promoter that is preferentially active in lymphocytes. The isoforms generated by this alternative promoter have no amino-terminal beta-catenin binding domain and may function in a dominant negative manner (6-8).
Applications:
Suitable for use in Western Blot, Flow Cytometry, Immunofluorescence. Other applications not tested.
Recommended Dilution:
Western Blot: 1:1000. Incubate membrane with diluted antibody in 1X TBS, 5% BSA, 0.1% Tween-20 at 4ºC with gentle shaking, overnight.
Flow Cytometry: 1:400
Immunofluorescence (IF-IC): 1:200
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
