Luciferase is a luminescent enzyme produced by Photinus pyralis, also referred to as the common eastern firefly or North American firefly. It is known for its role as a reporter protein and can be used to quantify expression levels from promoters in tissue culture and in vivo. Luciferase catalyses the oxygenation of luciferin using ATP and oxygen to produce oxyluciferin, a highly unstable, singlet-excited compound that emits light upon relaxation to its ground state. Bioluminescence assay systems have become increasingly used in biology and medical research laboratories in addition to (or as alternatives to) fluorescence and chemiluminescence detection strategies. Luciferase enzymes isolated from different animal species have inherent variability in light emission, allowing two or more luciferase enzymes to be used in combination for multiplex analyses, including in vivo imaging, cell viability and single and dual-spectral luciferase reporter assays. Applications: Suitable for use in Western Blot, Immunohistochemistry and Immunofluorescence. Other applications have not been tested. Recommended Dilutions: Western Blot: 1-3ug/ml. Observed in mouse heart, liver, kidney, and cortex tissues containing added purified luciferase protein. Immunohistochemistry (Frozen): 15-50ug/ml. Detects tissue specific expression of Luciferase protein in Drosophila embryos using Gal4-UAS system with two different Gal4 drivers. Immunofluorescence: 10-30ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
