Luciferase from the firefly has become one of the more widely used reporter proteins for the study of gene expression. Luciferase catalyzes a bioluminescent reaction which requires the substrate luciferin as well as Mg2+ and ATP. Mixing these reagents with the cell extract containing luciferase, results in a flash of light that decays rapidly. This light can be detected by a luminometer. The total light emission is proportional to the luciferase activity of the sample Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 0.5-2ug/ml detects 5ng of recombinant luciferase that was added to 3T3 cell lysate. 3T3 cell lysate containing 5ng of recombinant luciferase was resolved by electrophoresis, transferred to nitrocellulose and probed with L5999-04. Proteins were visualized using an IgG (HRP) goat anti-mouse and a chemiluminescence detection system. Immunohistochemistry: This antibody has been used by an independent laboratory for qualitative and quantitative assessment of gene expression systems employing luciferase reporter constructs in Drosophila embryos. Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt x Mo I1904-06H: IgG, H&L (HRP) (X-Adsorbed) Pab Gt x Mo Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
