Peutz-Jeghers syndrome (PJS) is caused by mutation of a widely expressed protein kinase termed LKB1 that is predicted to function as a tumor suppressor. LKB1 is a 433aa protein possessing a kinase domain only distantly related to other mammalian kinases. The amino-terminal noncatalytic domain comprises both a nuclear localization signal and a putative cytoplasmic retention signal. The mechanism by which LKB1 is regulated in cells is not known, nor have any of its physiological substrates been identified. Recent studies have demonstrated that LKB1 is phosphorylated in cells at serines 31, 325 and 431, and at threonines 189, 336 and 366. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000, incubate membrane with diluted primary antibody in TBS, 5% BSA, 0.1% Tween 20 at 4°C with gentle shaking, overnight. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
