Interferon regulatory factor 3 (IRF3) is a 50 kD, 427aa, member of the IRF family of proteins. Human IRF3 is a transcription factor that binds the interferon-sensitive response element (ISRE) and is activated by Toll-like receptors, TLR3 and TLR4. Viral entry or engagement of TLR3 or TLR4 stimulates IRF3 phosphorylation, nuclear translocation and stimulation of IFN production. Applications: Suitable for use in Western Blot, Flow Cytometry, Immunocytochemistry and ELISA. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml (w/ appropriate secondary reagents). Detection limit is ~10ng/lane (non-reducing) and ~1ng/lane (reducing). Flow Cytometry: 50ug/ml and add 10ul of the diluted solution to 1-2.5x10e5 HeLa cells in a total reaction volume ≥200ul. The binding of unlabeled antibodies may be visualized by adding 10ul of a 25ug/ml stock solution of a secondary developing reagent such as anti-goat IgG (FITC) Immunocytochemistry: 10ug/ml detects human IRF3 in HeLa cells. Cells were fixed with PBS, 4% paraformaldehyde for 20 min at RT and blocked with PBS, 10% normal donkey serum, 0.1% Triton X-100, and 1% BSA for 45 min at RT. After blocking, cells were incubated with diluted primary antibody overnight at 4° C followed by fluorochrome-coupled anti-goat IgG at RT in the dark for one hour. Between each step, cells were washed with PBS, 0.1% BSA. ELISA: 0.5-1.0ug/ml (w/ appropriate secondary reagents), detection limit is ~0.2 ng/well. Optimal dilutions to be determined by researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute by adding sterile 40-50% glycerol, PBS. Aliquot and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Selected for its ability to recognize human IRF3 in the applications listed below. In direct ELISA and Western Blots, this antibody shows ~60% crossreactivity with rmIRF3. Species Sequence Homology: mouse-76% and porcine-83%.
