The NF-kappaB/Rel transcription factors are present in the cytosol in an inactive state, complexed with the inhibitory IkappaB proteins. Most agents that activate NF-kappaB do so through a common pathway based on phosphorylation- induced, proteasome-mediated degradation of IkappaB. The key regulatory step in this pathway involves activation of a high molecular weight IkappaB kinase (IKK) complex, whose catalysis is generally carried out by three tightly associated IKK subunits. IKK alpha and IKK beta serve as the catalytic subunit of the kinase. IKK gamma serves as it regulatory subunit. Activation of IKK depends on phosphorylation. The serines 177 and 181 in the activation loop of IKK beta (176 and 180 in IKK alpha) are the specific sites whose phosphorylation causes a conformational change that results in kinase activation.
Applications:
Suitable for use in ELISA, Western Blot, Immunoprecipitation and Immunocytochemistry. Other applications not tested.

Immunocytochemistry (ABC): 1:200

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
