The histone tail acetylation promotes transcriptional activation. The identification of histone acetyltransferases (HATs) and their large multiprotein complexes has yielded important insights into how these enzymes regulate transcription (1,2). HAT complexes interact with sequence-specific activator proteins and target specific genes. In addition to histones, some HATs can acetylate nonhistone proteins, suggesting multiple roles for these enzymes (3). In another respect, histone deacetylation is involved in silencing of gene transcription (4). Mammalian histone deacetylases can be divided into two classes on the basis of their similarity to various yeast histone deacetylases (5). The first class is represented by its closeness to the yeast Rpd3- like proteins (HDAC 1, 2, 3 and 8), and the second class, which has similarities to yeast Hda1-like proteins (HDAC 4, 5, 6 and 7). Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
