Granzyme A, also known as CTLA-3, is the most abundant serine protease found specifically in the cytotoxic granules of cytotoxic T lymphocytes and NK cells. It is released during cellular activation and enters target cells through perforin channels. Granzyme A mediates target cell killing inducing caspase-independent cell death. Human Granzyme A is synthesized as a precursor with a signal peptide, a two amino acid propeptide, and a mature chain. Mature human Granzyme A shares 71% amino acid sequence identity with mouse Granzyme A. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry:10ul labels 5x10e5 cells in a total reaction volume ≤200ul. For intracellular staining, fix cells with 4% paraformaldehyde and permeabilize with 0.1% saponin in PBS. Following a 30 min incubation with G8956-05, wash cells with 0.1% saponin. Optimal dilutions to be determined by the researcher. Hybridoma: NSO myeloma cells with spleen cells from Balb/c mice. Storage and Stability: May be stored at 4°C before opening. DO NOT FREEZE! Stable at 4°C as an undiluted liquid. Stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Freezing Allophycocyanin conjugates will result in a substantial loss of enzymatic activity.
