Fibroblast growth factor receptor substrate 2 (FRS2, also called Suc-associated neurotrophic factor-induced tyrosine -phosphorylated target or SNT) participates in the transmission of extracellular signals from the fibroblast growth factor receptor (FGFR). Activation of the FGFR leads to tyrosyl phosphorylation of FRS2. Two FRS2 family members have been identified: FRS2-alpha (SNT1) and FRS2-beta (SNT2), which can directly bind to a range of activated receptor tyrosine kinases (RTKs) and become the substrate for these RTKs. Once they are tyrosyl phosphorylated, they can recruit SH2 domain-containing proteins including Grb2 and SHP-2, mediating a downstream biochemical and biological response. Tyrosine 436 was shown to be required for efficient SHP-2 recruitment, whereas tyrosine 196 functions as docking sites for Grb2-Sos complexes.
Applications:
Suitable for use in Western Blot. Other applications not tested.
Recommended Dilution:
Western Blot: 1:1000, incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight.
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
