Alpha-fetoprotein (AFP) is a 65kD glycoprotein found in the serum of the mammalian fetal liver, yolk sac and GI tract. Expression of AFP in adult cells is low. However, synthesis aberrantly occurs in adult liver cancer cells (1,2). The tumor suppressor gene p53 and beta-catenin are both involved in the regulation of AFP expression. In normal adult cells p53 binds to the repressor region of the AFP gene, thereby blocking transcription. Mutations in both p53 and beta-catenin are associated with aberrant expression of AFP. Elevated serum levels are predictive of hepatocellular carcinoma (3). Applications: Suitable for use in Flow Cytometry, Immunofluorescence (IF-IC) and Western Blot. Other applications not tested. Recommended Dilution: Immunofluorescence (IF-IC): 1:100 Western Blot: 1:1000 Flow Cytometry: 1:200 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
