The ezrin, radixin and moesin (ERM) proteins function as linkers between the plasma membrane and the actin cytoskeleton, and are involved in cell adhesion, membrane ruffling and microvilli formation (1). ERM proteins also undergo intra-or intermolecular interaction between the N-and C-terminal domains, which makes them exist in inactive, cytosolic monomers or dimers (2). Phosphorylation at a C-terminal threonine residue (Thr567 of ezrin, Thr564 of radixin, Thr558 of moesin), which disrupts the N-and C-terminal association, may play a key role in modulating the conformation and function of ERM proteins (3,4). It has also been demonstrated that phosphorylation at Thr567 of ezrin is required for cytoskeletal rearrangements and oncogene-induced transformation (5). Ezrin is also phosphorylated at tyrosine residues upon growth factor stimulation. Phosphorylation of Tyr353 of ezrin transmits a survival signal during epithelial differentiation (6). Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
