Estrogen receptor alpha (ER alpha), a member of the steroid receptor superfamily, contains highly conserved DNA binding
(DBD) and ligand binding domains (LBD) (1). Through its estrogen-independent and estrogen- dependent activation domains (AF-1 and AF-2, respectively), ER alpha regulates transcription by recruiting coactivator proteins and interacting with general transcriptional machinery (2). Phosphorylation provides an important mechanism to regulate ER alpha activity (3,4). ER alpha is phosphorylated on multiple sites (5). Serines 104 and 106 are located in the N-terminal transcription activation function domain AF-1, and phosphorylation of Ser104/106 plays an important role in regulating ER alpha activity (5).
Applications:
Suitable for use in ELISA, Western Blot, Immunocytochemistry and Immunofluorescence.
Other applications not tested.
Recommended Dilution:
Western Blot: 1:1000
Immunofluorescence: 1:50
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
