The nucleotide excision repair (NER) pathway has evolved to deal with UV-induced DNA lesions and also protects against a broad range of endogenously generated bulky DNA adducts. Ercc1 is essential for NER where it acts in a complex with Xpf (Ercc4) to make the incision 5' to the lesion site. The protein ERCC1 is also involved in recombination repair pathway. It is preferentially expressed in both neural and haematopoietic stem cells. It has a size of 15kb and is located on human chromosome 19. It exhibits homology to the yeast RAD10 repair protein and its longer C-terminus displays similarity to parts of the E. coli repair proteins uvrA and uvrC. The pathological role of ERCC1 has been observed in COFS syndrome type 4. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: ELISA: 1:64,000 Western Blot: 0.3-1ug/ml, observed in lysates of cell lines A431 and Kelly on ~38kD bands Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
