CD200 R1, also known as OX-2 receptor, is one of four CD200 R related proteins within the immunoglobulin superfamily. CD200 R1 is a type I transmembrane protein with a PTB-signaling motif in its cytoplasmic domain. Engagement of CD200 R1 by CD200 elicits an inhibitory signal in mast cells, basophils, macrophages, and dendritic cells. Alternate splicing of the human CD200 R1 mRNA generates four isoforms, two of which are truncated in the Ig-C2 domain and are likely secreted. Within the ECD, human CD200 R1 shares 56% aa sequence identity with mouse and rat CD200 R1. Applications: Suitable for use in Western Blot, Flow Cytometry, ELISA and Neutralization. Other applications not tested. Recommended Dilution: Western Blot: 0.1ug/ml Flow Cytometry: 0.25ug per 10e6 cells. Tested on human immature dendritic cells differentiated from human peripheral blood mononuclear cell derived CD14+ cells treated with 20ng/ml recombinant human IL-4 and 50ng recombinant human GM-CSF for 6-7days. Neutralization: 3-15ug/ml blocks 50% of the binding of 2.5ng/ml of recombinant human CD200/Fc to immobilized recombinant human CD200 Fc Chimera to immobilized recombinant human CD200 R1 Fc Chimera coated at 2ug/ml (100ul/well). 50ug/ml of this antibody blocks >90% of the binding. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
