Bromodeoxyuridine (BrdU) is a thymidine analog and is specifically incorporated into DNA during DNA synthesis. Detection of bromodeoxyuridine (BrdU) incorporated into DNA is a powerful tool to study the cytokinetics of normal and neoplastic cells. In vitro or in vivo labeling of tumor cells with the thymidine analogue BrdU and the subsequent detection of incorporated BrdU with specific anti-BrdU monoclonal antibodies is an accurate and comprehensive method to quantitate the degree of DNA-synthesis. BrdU is incorporated into the newly synthezised DNA of the S-phase cells and can thus provide an estimate for the fraction of cells in S-phase. Also dynamic proliferative information (such as the S-phase transit rate and the potential doubling time) can be obtained, by means of bivariate BrdU/DNA flow cytometric analysis. Applications: Suitable for use in ELISA, Western Blot, Immunofluorescence, Immunohistochemistry, Immunoprecipitation and Fluorescent In Situ Hybridization (FISH) Assay. Other applications not tested. Recommended Dilutions: Western Blot: Band is observed at ~307kD Immunohistochemistry: Frozen and paraffin sections. Enzymatic antigen retrieval and DNA acid denaturation required. Fluorescent In Situ Hybridization: 1:25; Use a rabbit anti-sheep AMCA secondary antibody. Note: Denature DNA sample first so base is accessible to BrdU antibody. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
