Bcl 2 has the ability to exert a survival function in response to a wide range of apoptotic stimuli by blocking mitochondrial release of cytochrome c (1). It has been implicated in modulating mitochondrial Ca2+ homeostasis and proton flux (2). Several phosphorylation sites have been identified within the Bcl 2 loop domain, including serine 70 (3). It has been suggested that phosphorylation at Ser70 may involve an ASK1/MKK7/JNK1 pathway and may be a marker for mitotic events (4,5). Interleukin 3 and JNK induced Bcl 2 phosphorylation at Ser70 may be required for its enhanced antiapoptotic functions (6). Applications: Suitable for use in ELISA, Immunofluorescence, Flow Cytometry and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000. Incubate membrane with diluted antibody in 5% w/v BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight. Western blot analysis of extracts from Jurkat cells, untreated or treated with paclitaxel (1uM, overnight) and with or without λphosphatase, using phospho-Bcl-2 (Ser70) Rabbit mAb (upper) or Bcl-2 (lower). Flow Cytometry: 1:25. Flow cytometric analysis of Jurkat cells, using Phospho-Bcl-2 (Ser70) (5H2) Rabbit mAb versus propidium iodide (DNA content). Immunofluorescence (Immunocytochemistry): 1:200. Immunofluorescent analysis of MCF-7 cells with or without treatment of paclitaxel (1uM, overnight), using phospho-Bcl-2 (Ser70) Rabbit mAb. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
