Bad is a proapoptotic member of the Bcl-2 family that can displace Bax from binding to Bcl-2 and Bcl-xL, resulting in cell death. Survival factors such as IL-3 can inhibit the apoptotic activity of Bad by activating intracellular signaling pathways that result in the phosphorylation of Bad at Ser112 and Ser136. Phosphorylation at these sites results in the binding of Bad to 14-3-3 proteins and the inhibition of Bad binding to Bcl-2 and Bcl-xL. Akt has been shown to promote cell survival via its ability to phosphorylate Bad at Ser136. Ser112 has been shown to be the substrate in vivo and in vitro of p90RSK and mitochondria-anchored PKA. Phosphorylation of Ser155 by PKA plays a critical role in blocking the dimerization of Bad to Bcl-xL because of its position in the BH3 domain.
Applications:
Suitable for use in Western Blot. Other applications not tested.
Recommended Dilution:
Western Blot: 1:500. Diluted antibody in TBS, 5% BSA, 0.1% Tween-20. Incubate at 4°C overnight with gentle shaking.
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
