Cleavage of amyloid precursor protein (APP) into 40 or 42 amino acid amyloid beta (ab) peptides is a result of the action of two proteases beta- and gamma-secretase. The beta-site APP cleaving enzyme, or BACE1, is a transmembrane aspartic protease that is localized in the golgi and endosomes. Low levels of BACE1 mRNA are found in most adult tissues with a higher level of expression in pancreas. Applications: Suitable for use in ELISA, Immunoprecipitation and Neutralization. Other applications not tested. Recommended Dilutions: ELISA: Use in a Direct ELISA. Immunoprecipitation: 25ug/ml to immunoprecipitate BACE-1 from conditioned media of cell culture spike with recombinant human BACE1 or recombinant mouse BACE1. Western Blot after Immunoprecipitation. Neutralization: The Neutralization Dose (ND50) is typically 10ug/ml. It is measured by the ability of the antibody to neutralize recombinant human BACE1 or recombinant mouse BACE1 cleavage of the fluorogenic peptide substrate Mca-SEVNLDAEFRK(Dnp)RR-NH2 (10uM). Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months after receipt at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
