Aurora A/AIK, a cell cycle-regulated serine/threonine protein kinase, is overexpressed in many tumor cell lines. Phosphorylation of threonine 288, which is within the activation loop of the kinase, results in a significant increase in its enzymatic activity. Aurora A/AIK is regulated by proteasome-dependent degradation, and phosphorylation on Thr288 may target it for degradation during mitosis. Thus, phosphorylation of Aurora A/AIK at Thr288 is important for both kinase activity and stability. Applications: Suitable for use in ELISA, and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1,000. Incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
