ADAM12 is a 120kD type I transmembrane glycoprotein member of the metzincin family of proteases. It is synthesized as a 909aa preproprecursor that contains a 25kD proregion (aa29-207) and a 92kD mature molecule. Proteolytic cleavage in the Golgi removes the propeptide prior to cell-surface expression. A 68kD soluble splice form exists in pregnancy where 36aa replace the C-terminal 205aa (705-909). This circulating ADAM12 isoform remains associated with its prodomain via noncovalent linkage, possibly acting as an enzyme regulator. Over aa29-513, human ADAM12 shares 78% and 79% aa identity with bovine and mouse ADAM12, respectively. Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: ELISA: 1:64,000 Western Blot: 0.1-0.3ug/ml, observed in human heart lysates on ~90kD bands Immunohistochemistry (Paraffin): 3-5ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
