Recognizes a protein of 40kD, identified as Thyroid transcription factor-1 (TTF-1). TTF-1 is a member of the NKx2 family of homeodomain transcription factors. It is expressed in epithelial cells of the thyroid gland and the lung. Nuclei from liver, stomach, pancreas, small intestine, colon, kidney, breast, skin, testes, pituitary, prostate, and adrenal glands are unreactive. Anti-TTF-1 is useful in differentiating primary adenocarcinoma of the lung from metastatic carcinomas originating in the breast, mediastinal germ cell tumors, and malignant mesothelioma. It can also be used to differentiate small cell lung carcinoma from lymphoid infiltrates. Loss of TTF-1 expression in non-small cell lung carcinoma has been associated with aggressive behavior of such neoplasms. TTF-1 reactivity is also seen in thyroid malignancies. Applications: Suitable for use in Flow Cytometry, Immunofluorescence, Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 5-10ul/10e6 cells Immunofluorescence: 1:50-1:100 Western Blot: 1:100-1:200 Immunohistochemistry (Frozen & Formalin fixed paraffin embedded): 1:100-1:200/30min @ RT Immunoprecipitation: 5-10ul/500ug protein lysate Optimal dilutions to be determined by the researcher. Positive Control: MAD109, MLE-15, H441-4, or H345 cells. Normal thyroid or lung. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
