Expression of DOG-1 protein is elevated in the gastrointestinal stromal tumors (GIST's), c-kit signaling-driven mesenchymal tumors of the GI tract. DOG-1 is rarely expressed in other soft tissue tumors, which, due to appearance, may be difficult to diagnose. Immunoreactivity for DOG-1 has been reported in 97.8% of scorable GIST's, including all c-kit negative GIST's. Overexpression of DOG-1 has been suggested to aid in the identification of GISTs, including Platelet-Derived Growth Factor Receptor Alpha (PDGFR alpha) mutants that fail to express c-kit antigen. The overall sensitivity of DOG1 and c-kit in GIST's is nearly identical: 94.4% vs. 94.7%. Applications: Suitable for use in Flow Cytometry, Western Blot, Immunofluorescence, Immunoprecipitation and Immunohistochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 5-10ul/10e6 cells Western Blot: 1:100-1:200 Immunofluorescence: 1:50-1:100 Immunoprecipitation: 5-10ul/500ug protein lysate Immunohistochemistry (Formalin fixed paraffin embedded): 1:100-1:200/30min @ RT Optimal dilutions to be determined by the researcher. Positive Control: Gastrointestinal Stromal Tumor (GIST) or testicular germ cell tumor. Melanocytes in the basal layer of the epidermis and mast cells in the derm is of normal skin. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
