Sclerostin is a secreted glycoprotein with a C-terminal cysteine knot-like (CTCK) domain and sequence similarity to the DAN (differential screening-selected gene aberrative in neuroblastoma) family of bone morphogenetic protein (BMP) antagonists. Loss-of-function mutations in this gene are associated with an autosomal-recessive disorder, sclerosteosis, which causes progressive bone overgrowth. A deletion downstream of this gene, which causes reduced sclerostin expression, is associated with a milder form of the disorder called van Buchem disease. Applications: Suitable for use in Western Blot, Immunohistochemistry and ELISA. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Immunohistochemistry (FFPE): 1:10-1:50 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Immunohistochemistry analysis in formalin-fixed and paraffin-embedded human breast carcinoma tissue reacted with 042148, which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of 042148 for immunohistochemistry.

Immunohistochemistry (FFPE) analysis staining of SOST in human liver tissue sections using 042148. Tissue was fixed with formaldehyde and blocked with 3% BSA for 0.5 hour at RT; antigen retrieval was by heat mediation with a citrate buffer (pH 6). Samples were incubated with 042148 (1:25) for 1 hours at 37°C. A undiluted biotinylated goat polyvalent antibody was used as the secondary antibody.
