The Mcl-1 protein belongs to the Bcl-2 family. Alternative splicing occurs at this locus and two transcript variants encoding distinct isoforms have been identified. The longer gene product (isoform 1) enhances cell survival by inhibiting apoptosis while the alternatively spliced shorter gene product (isoform 2) promotes apoptosis and is death-inducing. Applications: Suitable for use in ELISA, Western Blot, Immunohistochemistry and Flow Cytometry. Other applications have not been tested. Recommended Dilutions: Western Blot: 1:2000 Immunohistochemistry (FFPE): 1:10-1:50 Flow Cytometry: 1:25 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Overlay histogram showing A431 cells stained with 038275 (green line). The cells were fixed with 2% paraformaldehyde (10mins) and then permeabilized with 90% methanol for 10mins. The cells were then incubated in 2% BSA to block non-specific protein-protein interactions followed by 038275 (1:25) for 60mins at 37ºC. The secondary antibody used was Goat-Anti-Rabbit IgG, DyLight® 488 Conjugated Highly Cross-Adsorbed (1:200) for 40mins at 37ºC. Isotype control antibody (blue line) was rabbit IgG (1ug/1x10e6 cells) used under the same conditions. Acquisition of >10,000 events was performed.
