The Mcl-1 protein belongs to the Bcl-2 family. Alternative splicing occurs at this locus and two transcript variants encoding distinct isoforms have been identified. The longer gene product (isoform 1) enhances cell survival by inhibiting apoptosis while the alternatively spliced shorter gene product (isoform 2) promotes apoptosis and is death-inducing. Applications: Suitable for use in Western Blot, Immunohistochemistry, Flow Cytometry and FLISA. Other applications have not been tested. Recommended Dilutions: Immunohistochemistry: Formalin-fixed, paraffin-embedded sections Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.

Overlay histogram showing A431 cells stained with 038275 (green line). The cells were fixed with 2% paraformaldehyde (10mins) and then permeabilized with 90% methanol for 10mins. The cells were then incubated in 2% BSA to block non-specific protein-protein interactions followed by 038275 (1:25) for 60mins at 37ºC. The secondary antibody used was Goat-Anti-Rabbit IgG, DyLight® 488 Conjugated Highly Cross-Adsorbed (1:200) for 40mins at 37ºC. Isotype control antibody (blue line) was rabbit IgG (1ug/1x10e6 cells) used under the same conditions. Acquisition of >10,000 events was performed.
