The alpha (HBA) and beta (HBB) loci determine the structure of the 2 types of polypeptide chains in adult hemoglobin, Hb A. The normal adult hemoglobin tetramer consists of two alpha chains and two beta chains. Mutant beta globin causes sickle cell anemia. Absence of beta chain causes beta-zero-thalassemia. Reduced amounts of detectable beta globin causes beta-plus-thalassemia. The order of the genes in the beta-globin cluster is 5'-epsilon -- gamma-G -- gamma-A -- delta -- beta--3'. Applications: Suitable for use in ELISA, Western Blot, Immunohistochemistry and Flow Cytometry. Othre applications not tested. Recommended Dilutions: Western Blot: 1:2000 Immunohistochemistry (FFPE): 1:10-1:50 Flow Cytometry: 1:10-1:50 Optimal dilutions to be determined by the researcher Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Flow Cytometric analysis of K562 cells using 036445 (right histogram) compared to a negative control cell (left histogram). FITC-conjugated goat-anti-rabbit secondary antibodies were used for the analysis.

Overlay histogram showing K562 cells stained with 036445 (green line). The cells were fixed with 4% paraformaldehyde (10mins) and then permeabilized with 90% methanol for 10mins. The cells were then incubated in 2% BSA to block non-specific protein-protein interactions followed by 036445 (1:25) for 60mins at 37ºC. The secondary antibody used was Alexa Fluor® 488 goat anti-rabbit lgG (H+L) at 1:400 for 40mins at 37ºC. Isotype control antibody (blue line) was rabbit IgG1 (1ug/1x10e6 cells) used under the same conditions. Acquisition of >10,000 events was performed.
