HBA2 located on chromosome 16 spans about 30 kb and includes seven loci: 5'- zeta - pseudozeta - mu - pseudoalpha-1 - alpha-2 - alpha-1 - theta - 3'. The alpha-2 (HBA2) and alpha-1 (HBA1) coding sequences are identical. These genes differ slightly over the 5' untranslated regions and the introns, but they differ significantly over the 3' untranslated regions. Two alpha chains plus two beta chains constitute HbA, which in normal adult life comprises about 97% of the total hemoglobin; alpha chains combine with delta chains to constitute HbA-2, which with HbF (fetal hemoglobin) makes up the remaining 3% of adult hemoglobin. Alpha thalassemias result from deletions of each of the alpha genes as well as deletions of both HBA2 and HBA1; some nondeletion alpha thalassemias have also been reported. Applications: Suitable for use in Flow Cytometry, Western Blot, Immunohistochemistry and FLISA. Other applications have not been tested. Recommended Dilution: Immunohistochemistry: Formalin-fixed, paraffin-embedded sections. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.

Overlay histogram showing K562 cells 036444 (green line). The cells were fixed with 2% paraformaldehyde (10mins) and then permeabilized with 90% methanol for 10mins. The cells were then incubated in 2% BSA to block non-specific protein-protein interactions followed by 036444 (1:25) for 60mins at 37ºC. The secondary antibody used was Goat-Anti-Rabbit IgG, DyLight® 488 Conjugated Highly Cross-Adsorbed at 1:200 for 40mins at 37ºC. Isotype control antibody (blue line) was rabbit IgG (1ug/1x10e6 cells) used under the same conditions. Acquisition of >10,000 events was performed.
