CES2 is a member of the carboxylesterase large family. The family members are responsible for the hydrolysis or transesterification of various xenobiotics, such as cocaine and heroin, and endogenous substrates with ester, thioester, or amide bonds. They may participate in fatty acyl and cholesterol ester metabolism, and may play a role in the blood-brain barrier system. The protein encoded by this gene is the major intestinal enzyme and functions in intestine drug clearance. Applications: Suitable for use in Western Blot, Immunohistochemistry, Flow Cytometry and ELISA. Other applications not tested. Recommended Dilutions: Western Blot: 1:1000 Immunohistochemistry (FFPE): 1:50-1:100 Flow Cytometry: 1:25 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Immunohistochemistry analysis in formalin fixed and paraffin embedded human hepatocarcinoma using 033793 followed by peroxidase conjugation of the secondary antibody and DAB staining. This data demonstrates the use of 033793 for immunohistochemistry.

Flow Cytometric overlay histogram showing U-87 MG cells stained with 033793 (green line). The cells were fixed with 2% paraformaldehyde (10 min) and then permeabilized with 90% methanol for 10 min. The cells were then icubated in 2% BSA to block non-specific protein-protein interactions followed by033793 (1:25) for 60 min at 37oC. The secondary antibody used was Alexa Fluor® 488 goat anti-rabbit lgG (H+L) at 1:400 dilution for 40 min at 37oC. Isotype control antibody (blue line) was rabbit IgG1 (1ug/1x10e6 cells) used under the same conditions. Acquisition of >10, 000 events was performed.
