The protein encoded by this gene is a signal transducer shared by many cytokines, including interleukin 6 (IL6), ciliary neurotrophic factor (CNTF), leukemia inhibitory factor (LIF), and oncostatin M (OSM). This protein functions as a part of the cytokine receptor complex. The activation of this protein is dependent upon the binding of cytokines to their receptors. vIL6, a protein related to IL6 and encoded by the Kaposi sarcoma-associated Herpes virus, can bypass the interleukin 6 receptor (IL6R) and directly activate this protein. Knockout studies in mice suggest that this gene plays a critical role in regulating myocyte apoptosis. Alternatively spliced transcript variants encoding distinct isoforms have been described. A related pseudogene has been identified on chromosome 17. Applications: Suitable for use in Western Blot, Immunohistochemistry and FLISA. Other applications have not been tested. Recommended Dilution: Immunohistochemistry: Formalin-fixed, paraffin-embedded sections Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.

Staining CD130 in Human skeletal muscle tissue sections by Immunohistochemistry (FFPE) using 033450. Tissue was fixed with formaldehyde and blocked with 3% BSA for 0.5 hour at RT; antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with 033450 (1:25) for 1 hours at 37°C. A undiluted biotinylated goat polyvalent antibody was used as the secondary antibody.

Immunohistochemistry analysis in formalin fixed and paraffin embedded human uterus tissue using 033450 followed by peroxidase conjugation of the secondary antibody and DAB staining.This data demonstrates the use of 033450 for immunohistochemistry. Clinical relevance has not been evaluated.
