Desc1 :

ADM, a hypotensive peptide found in human pheochromocytoma, consists of 52 amino acids, has 1 intramolecular disulfide bond, and shows a slight homology with the calcitonin gene-related peptide. It may function as a hormone in circulation control because it is found in blood in a considerable concentration. The precursor, called preproadrenomedullin, is 185 amino acids long. By RNA-blot analysis, human adrenomedullin mRNA was found to be highly expressed in several tissues. Genomic ADM DNA consists of 4 exons and 3 introns, with the 5-prime flanking region containing TATA, CAAT, and GC boxes. Applications: Suitable for use in Western Blot, Flow Cytometry and ELISA. Other applications have not been tested. Recommended Dilutions: Western Blot: 1:1000-1:2000 Flow Cytometry: 1:10-1:50 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Overlay histogram showing A549 cells stained with 031644 (green line). The cells were fixed with 2% paraformaldehyde (10 min) and then permeabilized with 90% methanol for 10 min. The cells were then icubated in 2% BSA to block non-specific protein-protein interactions followed by 031644 (1:25) for 60 mins at 37ºC. The secondary antibody used was Goat-Anti-Rabbit IgG, DyLight® 488 Conjugated Highly Cross-Adsorbed at 1:400 for 40 mins at 37ºC. Isotype control antibody (blue line) was rabbit IgG (1ug/1x10e6 cells) used under the same conditions. Acquisition of >10, 000 events was performed.
