Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the 'beads on a string' structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine. Histone variant H3.3 is typically enriched in active chromatin. Applications: Suitable for use in Western Blot, Dot Blot and Chromatin Immunoprecipitation. Other applications not tested. Recommended Dilution: ChIP: ~2ug per chromatin immunoprecipitation (dependent upon biological context). Optimal dilutions to be determined by the researcher. Positive Control: Includes normal rabbit IgG and primers specific for human GAPDH coding region Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
