JLK 6 is an inhibitor of -secretase that selectively inhibits APP cleavage without affecting other -secretase-mediated pathways. Prevents recovery of A 40 and A 42 from HEK293 cell overexpressing wild-type or Swedish-mutated APP (IC50 ~ 30 u M) but displays no effect on Notch cleavage and Notch-mediated intracellular signaling. Displays no activity on BACE1, BACE2, -secretase, the proteosome or GSK3 .
