Phospho-p70 S6 kinase alpha is a protein encoded by the RPS6KB1 gene which is approximately 59,2 kDa. Phospho-p70 S6 kinase alpha is localised to the mitochondrion and cell junctions. It is involved in pathways such as the common cytokine receptor gamma-c family signalling pathway, regulation of lipid metabolism insulin signalling-generic cascades, RET signalling and mTOR signalling. This protein falls under the ribosomal S6 kinase family of serine/threonine kinases. It responds to mTOR signalling and is activated in response to growth factors and nutrients to promote protein synthesis, cell growth, cell cycle progression and cell proliferation. It regulates protein synthesis through phosphorylation of EIF4B, RPS6 and EEF2K, and contributes to cell survival by repressing the pro-apoptotic function of BAD. Phospho-p70 S6 kinase alpha is expressed in the cells of the eye, nervous system, muscle, heart and liver. Mutations in the RPS6KB1 gene result in diseases such as tuberous sclerosis which manifests as hamartoma formation in multiple organ systems and glioblastoma multiforme which is a central nervous system tumour of grade IV histological malignancy. STJ90374 was affinity-purified from rabbit antiserum by affinity-chromatography using epitope-specific immunogen. The synthesized peptide derived from human p70 S6 kinase alpha around the phosphorylation site of S447. This primary antibody specifically binds to endogenous p70 S6 kinase α protein which only binds about S447 when S447 is phosphorylated.
