A DNA sequence encoding the HSV-1 gH (P06477) (Met1-Ala792) was fused with a Strep II at the C-terminus., constructed the plasmid 1; A DNA sequence encoding the HSV-1 gL (P10185) (Met1-Leu224) was fused with a polyhistidine tag at the C-terminus, constructed the plasmid 2. The two plasmids were co-expressed and the HSV-1 (strain 17) gH & gL Heterodimer was purified. The purified protein was biotinylated in vitro.
