Application Note :
This monoclonal antibody contains TBS/1% BSA and is suitable for ELISA, immunohistochemistry, flow cytometry, and western blotting. Specific conditions for reactivity should be optimized by the end user. Expect a band approximately 23 kDa in size corresponding to CD3 protein by western blotting in the appropriate cell lysate or extract. Jurkat cell lysates can be used as a control for western and flow. For immunohistochemistry, samples should be formalin fixed and paraffin embedded. Deparaffinize slides using xylene or xylene alternatives and graded alcohols. Staining requires boiling of sections in 10 mM citrate buffer pH 6.0 for 10 min followed by cooling at RT for 20 min. CD3 is a membrane protein. Tonsil tissue can be used as a positive control.

Immunogen :
Anti-CD3 monoclonal antibody was produced by repeated immunizations with a synthetic peptide corresponding to amino acid residues 156-168 of the epsilon chain of human CD3 protein. The hybridoma was produced by fusing New Zealand White rabbit splenocytes and myeloma cells using conventional technology.

Background :
The CDE3 gene encodes the CD3-epsilon polypeptide (also known as CD3, T-cell surface glycoprotein CD3 epsilon chain, T-cell surface antigen T3/Leu-4 epsilon chain and CD3e) , which together with CD3-gamma, -delta and -zeta, and the T-cell receptor alpha/beta and gamma/delta heterodimers, forms the T-cell receptor-CD3 complex. This complex plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways. The genes encoding the epsilon, gamma and delta polypeptides are located in the same cluster on chromosome 11. The epsilon polypeptide plays an essential role in T-cell development. Defects in this gene cause immunodeficiency. CD3 antibody is ideal for Immunology and Stem Cell research.
