Application Note :
This monoclonal antibody contains TBS/1% BSA and is suitable for ELISA, immunohistochemistry and western blotting. Specific conditions for reactivity should be optimized by the end user. Expect a band approximately 67 kDa in size corresponding to Estrogen Receptor protein by western blotting in the appropriate cell lysate or extract. Use MCF-7 as a positive control. For immunohistochemistry, samples should be formalin fixed and paraffin embedded. Deparaffinize slides using xylene or xylene alternatives and graded alcohols. Staining requires boiling of sections in 10 mM citrate buffer pH 6.0 for 10 min followed by cooling at RT for 20 min. Estrogen Receptor is found in human breast carcinoma tissue as a nuclear protein.

Background :
The ESR1 gene encodes an estrogen receptor (also known as ER, estradiol receptor, ER-alpha and nuclear receptor subfamily 3 group A member 1), which is a ligand-activated transcription factor composed of several domains important for hormone binding, DNA binding, and activation of transcription. The protein localizes to the nucleus where it may form a homodimer or a heterodimer with estrogen receptor 2. Estrogen and its receptors are essential for sexual development and reproductive function, but also play a role in other tissues such as bone. Estrogen receptors are also involved in pathological processes including breast cancer, endometrial cancer, and osteoporosis. Alternative splicing results in several transcript variants, which differ in their 5' UTRs and use different promoters.
