Western Blot showing detection of Recombinant Human VEGF-165. 50ng of protein (Lane 1) was run on a 4-20% gel and transferred to 0.45 µm nitrocellulose. After blocking with 1% BSA-TTBS (p/n MB-013, diluted to 1X) 30 min at 20°C, Anti-VEGF-165 (RABBIT) Antibody Peroxidase Conjugate (p/n 209-403-B99) secondary antibody was used at 1:1000 in Blocking Buffer for Fluorescent Western Blotting (p/n MB-070) and imaged using the Bio-Rad VersaDoc® 4000 MP. Arrow indicates correct 19 kDa molecular weight position expected for rH-VEGF-165.

Rockland's anti-Human VEGF-165 antibody shows detection of a band ~22 kDa in size corresponding to recombinant human VEGF-165. This isoform of VEGF tends to form a disulfide linked homodimer. Molecular weight markers are also shown (left). After transfer, the membrane was blocked overnight with 3% BSA in TBS followed by reaction with primary antibody at a 1:1,000 dilution. Detection occurred using peroxidase conjugated anti-Rabbit IgG (p/n 611-103-122) secondary antibody diluted 1:40,000 in blocking buffer (p/n MB-070) for 30 min at RT followed by reaction with FemtoMax™ chemiluminescent substrate. Image was captured using VersaDoc™ MP 4000 imaging system (Bio-Rad).

Anti-human VEGF-121 by western blot shows detection of recombinant Human VEGF-121 raised in E.coli. Recombinant (0.1 µg, 28.4 kDa) protein was loaded onto and resolved by SDS-PAGE, then transferred to nitrocellulose. The membrane was blocked with 1% BSA in TBST for 30 min at RT, followed by incubation with Rockland's, Inc. Anti-Human VEGF-121. After washing, membrane was probed with secondary antibody Dylight™ 649 Conjugated Anti-Rabbit IgG (H&L) (Goat) Antibody (611-143-122) diluted 1:20,000 in blocking buffer (p/n MB-070) for 30 min. at RT. Data was collected using Bio-Rad VersaDoc® 4000 MP imaging system.
