Purity and Specificity :

Anti-Parkin pS378 antibody is directed against human Parkin protein. The antibody was prepared from monospecific antiserum by immunoaffinity chromatography using phospho peptide coupled to agarose beads followed by solid phase adsorption(s) against non-phospho peptide and non-specific peptide to remove any unwanted reactivities. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Rabbit Serum. Immunolabeling of the parkin band is absent in parkin S378 mutants. This antibody is directed against human parkin proteins phosphorylated at Ser378. Cross reactivity with Parkin from other species has not been determined.

Background :

Parkin pS378 Antibody detects Parkin pS378 protein. Parkin is an E3 ligase in the ubiquitin-proteasome system. Hereditary Parkinson’s disease is most commonly caused by mutations in the parkin gene and is characterized by the progressive loss of dopaminergic neurons and the presence of Lewy bodies in the substania nigra. Recent evidence suggests that phosphorylation of parkin at Ser378 may have an important regulatory role on its E3 ubiquitin ligase activity.
