Immunohistochemistry of Rabbit Anti-Scp3 Antibody.
Tissue: Mouse Testis.
Antigen Retrieval: HIER using Citrate buffer for 20 min.
Fixative: none.
Primary Antibody: Anti-Scp3 at 1:100 for 30min at RT.
Secondary Antibody: Anti-Rabbit Poly-HRP-IgG Ready to Use for 8 min at RT.
CounterStain: Hematoxylin.
Substrate: DAB.
Analysis Results: Scp3 showed weak to moderate nuclear staining of spermatocytes. Other germ cells did not stain. The specificity of the nuclear staining is consistent with Scp3 staining.

Immunogen :
Anti-Scp3 antibody was prepared from whole rabbit serum produced by repeated immunizations with a synthetic peptide corresponding to a near N-terminal region of mouse Scp3 conjugated to Keyhole Limpet Hemocyanin (KLH).
Packing Type :
Dry Ice
Physical State :
Liquid (sterile filtered)
Preservative :
0.01% (w/v) Sodium Azide
Purity and Specificity :
This affinity purified antibody is directed against mouse Scp3. This product was affinity purified from monospecific antiserum by immunoaffinity purification. A BLAST analysis was used to suggest cross-reactivity with the antigen based on 100% homology with the immunizing sequence to mouse and golden hamster and 93% to rat.
Storage :
Store vial at -20° C or below prior to opening. This vial contains a relatively low volume of reagent (25 µL). To minimize loss of volume dilute 1:10 by adding 225 µL of the buffer stated above directly to the vial. Recap, mix thoroughly and briefly centrifuge to collect the volume at the bottom of the vial. Use this intermediate dilution when calculating final dilutions as recommended below. Store the vial at -20°C or below after dilution. Avoid cycles of freezing and thawing.
Synonyms :
Rabbit Anti-Synaptonemal complex protein 3 Antibody, SCP-3, Sycp3, Scp3, Synaptonemal complex protein 3
Target Species :
Mouse
Background :
Synaptonemal complex protein 3 (Scp3) is a component of the synaptonemal complexes (SCS), formed between homologous chromosomes during meiotic prophase. These proteins are required for centromere pairing during meiosis in male germ cells. It is required for normal meiosis during spermatogenesis and male fertility but plays a lesser role in female fertility. Required for efficient phosphorylation of HORMAD1 and HORMAD2. Mutations in this gene are associated with azoospermia in males and susceptibility to pregnancy loss in females. Anti-Scp3 Antibody is useful for researchers interested in Meiosis, Mitotic Cell Cycle, and fertility.
