TNF Induces phosphorylation of p65 in KBM-5 cells.  Cytoplasmic and nuclear protein lysates prepared after 0, 5, 10, 15, 30 and 60 minutes of 0.1 nM TNF treatment of KBM-5 cells shows inducible phosphorylation using phospho specific polyclonal anti-human pS276 p65.  Rockland Immunochemical's pan reactive anti p65 (code# 100-4165) was used a control to show the presence of total p65 in both the cytoplasmic and nuclear extracts.  Phosphorylation of p65 occurs after approximately 10 min of TNF exposure.  Migration of phosphorylated p65 into the nucleus occurs within a similar time frame.   HRP conjugated Gt-anti-Rabbit IgG was used to develop the western blot using a chemi-luminescent detection method.  Other detection methods will yield similar results. Rockland Immunochemical's Rabbit anti-p65 NLS was used at a 1:200 dilution to detect p65 in (A) control DU145 cells and (B) TNF stimulated DU145 cells.  Although DU145 show relatively high basal levels of nuclear p65 staining, significant enhancement of nuclear staining is seen in panel B as evidence of translocation and availability of the NLS to be bound by the antibody.  Cultured cells shown above were formalin-fixed.  Tissue staining (not shown) were formalin-fixed, paraffin embedded followed by citrate retrieval.  Blocking and hybridization included 5% NGS.

Rockland Monoclonal anti NFKB p65 (Rel A) antibody was used to detect p65 by immunofluorescence at a dilution of 1:5000. Hela cells were grown to sub-confluent on 18 mm2 glass coverslips #1.5. Cells were either unstimulated (A), or stimulated (B) with 50 ng/ml of TNF alpha for 30 min prior fixation. Cells were then fixed in methanol and blocked with 10% normal goat serum (NGS), in PBS, and TritonX 0.2% (Tx) and incubated for 1 hr at RT with primary ab, counterstained with DAPI and washed in PBS/NGS/Tx. Cells were incubated for 1 hr at RT with Atto 425 conjugated anti mouse secondary antibody for STED CW imaging. Data was collected on a STED-CW TCS-SP5 Confocal system equipped with a DFC 350FX camera allowing sequential acquisition in widefiled, confocal and STED CW imaging on the same system.

Rockland Affinity purified Anti-NFKB p65 (Rel A) pS276 600-401-264 lot 24040 was probed against Normal (Lane 1) and TNFalpha (Lane 2) Stimulated HeLa whole cell lysates. A band was observed between 55 and 72 kD corresponding to the expected MW of NFkB p65. The observed higher MW bands have not been characterized.
