Application Note :
This monoclonal antibody is suitable for ELISA, immunohistochemistry, flow cytometry, and western blotting. Specific conditions for reactivity should be optimized by the end user. Expect a band approximately 35 kDa in size corresponding to CD20 protein by western blotting in the appropriate cell lysate or extract. Ramos Cell lysate can be used as a positive control. CD20 is found in the cytoplasm and at the cell membrane. For immunohistochemistry, samples should be formalin fixed and paraffin embedded. Deparaffinize slides using xylene or xylene alternatives and graded alcohols. No special pretreatment of tissue is required for staining. Tonsil tissue or B cell lymphoma can be used as a positive control. For Flow cytometry use jurkat cell line as the positive control.

Anti-CD20 monoclonal antibody was produced by repeated immunizations with a synthetic peptide corresponding to amino acid residues at the carboxy terminus of human CD20 protein. The hybridoma was produced by fusing New Zealand White rabbit splenocytes and myeloma cells using conventional technology.

Background :
The MS4A1 gene encodes a member of the membrane-spanning 4A gene family termed the B-lymphocyte antigen CD20 (also known as CD20, membrane-spanning 4-domains subfamily A member 1, B-lymphocyte surface antigen B1, Leu-16 and Bp35). Members of this nascent protein family are characterized by common structural features and similar intron/exon splice boundaries and display unique expression patterns among hematopoietic cells and nonlymphoid tissues. The protein is expressed by B cells as a surface molecule which plays a role in the development and differentiation of B-cells into plasma cells. Monoclonal antibodies against CD20 are used to treat B-cell non-Hodgkin's lymphoma (Rituximab, Britumomab and Tositumomab). Ideal for Cancer, Immunology and Stem Cell research.
