Immunohistochemistry of Chicken Anti-Menin 1 Antibody. Tissue: Human Colon. Fixative: None. Antigen Retrieval: HIER using Citrate Buffer for 20mins. Primary Antibody: Anti-MEN1 at 1:100 for 30 in at RT. Secondary Antibody: Alpha Anti-Chicken IgG HRP at 1:500 for 20min at RT. Counter stain: Hematoxylin. Substrate: DAB. Analysis Report: MEN1 showed strong staining in a subset of colonic epithelial cells. MEN1 does not just stain neuroendocrine cells (1% of colonic cells) but also stains proliferating epithelial cells so this staining pattern was deemed physiologic.

This affinity purified antibody is directed against human Menin1.  This product is an IgY fraction antibody purified from monospecific chicken egg yolks by a multi-step process which includes selective precipitation and salt fractionation followed by extensive dialysis against the buffer stated above.  Blast analysis the immunogen sequence shows 100% identity with human, mouse, rat, bovine, canine, and is highly conserved.

Store vial at -20° C or below prior to opening. This vial contains a relatively low volume of reagent (25 µL). To minimize loss of volume dilute 1:10 by adding 225 µL of the buffer stated above directly to the vial. Recap, mix thoroughly and briefly centrifuge to collect the volume at the bottom of the vial. Use this intermediate dilution when calculating final dilutions as recommended below. Store the vial at -20°C or below after dilution. Avoid cycles of freezing and thawing.

Menin is a scaffold protein that functions in histone modification and epigenetic gene regulation. It is thought to regulate several pathways and processes by altering chromatin structure through the modification of histones. Essential component of a MLL/SET1 histone methyltransferase (HMT) complex, a complex that specifically methylates 'Lys-4' of histone H3 (H3K4). Functions as a transcriptional regulator. Binds to the TERT promoter and represses telomerase expression. Plays a role in TGFB1-mediated inhibition of cell-proliferation, possibly regulating SMAD3 transcriptional activity. Represses JUND-mediated transcriptional activation on AP1 sites, as well as that mediated by NFKB subunit RELA. Positively regulates HOXC8 and HOXC6 gene expression. May be involved in normal hematopoiesis through the activation of HOXA9 expression. May be involved in DNA repair.
