Wnt-9a in Human Breast Cancer Tissue. Wnt-9a was detected in immersion fixed paraffin-embedded sections of human breast cancer tissue using 15 ug/mL Mouse Anti-Human Wnt-9a Monoclonal Antibody (Catalog # MAB31571) overnight at 4 ░C. Before incubation with the primary antibody tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013 ). Detection of Human Wnt-9a by Western Blot. Western blot shows lysates of KATO-III human gastric carcinoma cell line and ME-180 human cervical epithelial carcinoma cell line. PVDF membrane was probed with 5 ╡g/mL Mouse Anti-Human Wnt-9a Monoclonal Antibody (Catalog # MAB3157) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007 ). For additional reference, recombinant human Wnt-9a (20 ng) was included. Specific bands for native Wnt-9a were detected at approximately 30 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 6 .
