Detection of Human Wnt-10b by Western Blot. Western blot shows lysates of SW480 human colorectal adenocarcinoma cell line and MDA-MB-468 human breast cancer cell line. PVDF membrane was probed with 0.5 ╡g/mL of Mouse Anti-Human Wnt-10b Monoclonal Antibody (Catalog # MAB7196) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018 ). A specific band was detected for Wnt-10b at approximately 50 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1 .

Wnt-10b in Human Breast Cancer Tissue. Wnt-10b was detected in immersion fixed paraffin-embedded sections of human breast cancer tissue using Mouse Anti-Human Wnt-10b Monoclonal Antibody (Catalog # MAB71961) at 15 ╡g/mL overnight at 4 ░C. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013 ). Tissue was stained using the Anti-Mouse HRP-DAB Cell Tissue Staining Kit (brown; Catalog # CTS002 ) and counter-stained with hematoxylin (blue).
