IL-8 Secretion Induced by Pam3CSK4 and Neutralization by Human TLR2 Antibody. The synthetic tripalmitoylated lipopeptide Pam3CSK4 stimulates IL-8 secretion in the HEK293 human embryonic kidney cell line transfected with human TLR2, in a dose-dependent manner (orange line), as measured by the Human CXCL8/IL-8 Quantikine ELISA Kit (Catalog # D8000C ). Detection of Mouse TLR2 by Western Blot. Western blot shows lysates of RAW 264.7 mouse monocyte/macrophage cell line. PVDF Membrane was probed with 1 ╡g/mL of Goat Anti-Mouse TLR2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1530) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019 ). A specific band was detected for TLR2 at approximately 100 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1 .
