Background :

Epidermal growth factor (EGF) has a profound effect on the differentiation of specific cells in vivo and is a potent mitogenic factor for a variety of cultured cells. The EGF precursor is believed to exist as a membrane-bound molecule which is proteolytically cleaved to generate the 53-amino acid peptide hormone that stimulates cells to divide. EGF exerts its actions by binding to the EGFR, a 170 kDa protein. Epidermal growth factor is also a key growth factor regulating cell survival. Through its binding to cell surface receptors, EGF activates an extensive network of signal transduction pathways that include activation of the PI3K/AKT, RAS/ERK and JAK/STAT pathways

Application Summary :

Neutralization:. To yield one-half maximal inhibition [ND50] of the biological activity of mEGF (0.1 ng/mL), a concentration of 1.0-1.5 g/mL of this antibody is required. ELISA:. In a sandwich ELISA (assuming 100 L/well), a concentration of 2.0-4.0 g/mL of this antibody will detect at least 100 pg/mL of recombinant murine EGF when used with our biotinylated antigen affinity purified anti-murine EGF as the detection antibody at a concentration of approximately 1.0-2.0 g/mL. Western Blot:. To detect mEGF by Western Blot analysis this antibody can be used at a concentration of 2.0-4.0 g/mL. Used in conjunction with compatible secondary reagents the detection limit for recombinant mEGF is 0.25-0.50 ng/lane under non-reducing conditions and 10.0-15.0 ng/lane under reducing conditions.

Storage :

EGF lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20 C. Reconstitute in sterile water to a concentration of 0.1 - 1.0 mg/mL. The antibody is stable for at least six weeks at 2-4 C when reconstituted.
