Background :
TGF beta 1 (TGFB1) is part of the transforming growth factor beta superfamily of cytokines. These play a role in activating the TGF beta signal transduction pathway. The TGF beta pathway is triggered by the binding of a ligand which brings together type I and type II receptor serine/threonine kinases on the surface of the cell. This then causes receptor II to phosphorylate the receptor I kinase domain. Subsequently the signal is multiplied via phosphorylation of the Smad proteins (PMID12809600). Transforming growth factor beta 1 is important in many aspects of proper cell function. However, TGF beta 1 has been shown to be a factor in many types of cancer. It has been demonstrated that via the TGF-B/Smad signaling pathway, TGF-B1 could stimulate epithelial to mesenchymal transition in squamous cell carcinoma of the head and neck cell lines (PMID: 21479366). Also, breast carcinoma MCF-7 cell growth was shown to be reduced in response to the down-regulation of expression of TGFB1 under hyperthermic conditions (PMID: 21455587). Furthermore, it has been suggested that overall survival of patients with non-small cell lung cancer treated with platinum-based chemotherapy could be predicted by SNPs in the TGF beta pathway (PMID: 21515830).
